What is HPLC purity?

⚡ Short answer

HPLC purity is the percentage of the detected signal that belongs to the main compound. HPLC separates a sample into its parts, and purity is the main peak's area divided by the total area of all peaks.

How HPLC works

A dissolved sample is pumped at high pressure through a column packed with fine particles. Different components travel through at different speeds, so they come out at different times.

A detector records each component as a peak on a graph called a chromatogram. For peptides the detector usually reads ultraviolet light at around 214 to 220 nm, where peptide bonds absorb.

How the percentage is worked out

Purity is the area of the main peak divided by the combined area of every peak, times 100. If the main peak is 99 units of area out of 100, the purity is 99%.

What HPLC purity does not tell you

Identity: HPLC alone cannot prove the main peak is the right compound. That is the job of mass spectrometry.
Quantity: purity says nothing about how many milligrams are in the vial.
Net peptide content: water and salts left from manufacturing do not show up as peaks, so a 99% pure powder is not 99% peptide by weight.
